RESUMO

The histological diagnosis of extranodal marginal zone lymphoma of the mucosa-associated lymphoid tissue (MALT lymphoma) is difficult for pathologists. Recently, digital pathology systems have been widely used to provide tools that can objectively measure lesions on slides. In this study, we measured the extent of marginal zone expansion in suspected MALT lymphoma cases and compared the results with those of a molecular clonality test. In total, 115 patients who underwent an IGH gene rearrangement test for suspected MALT lymphoma were included in this study. All cases were histologically classified into three patterns; "small lymphoid aggregates with no germinal center (Pattern 1)," "lymphoid follicles with germinal center (Pattern 2)" and " fused marginal zone or diffuse small lymphocytic proliferation (Pattern 3)." The proportions of monoclonality in Pattern 1, 2, and 3 were 25.0%, 55.0%, and 97.9%, respectively. The ratios of marginal zone thickness to germinal center diameter and entire lymphoid follicle area to germinal center area were measured in Pattern 2 cases using a digital pathology system. Combining the width cutoff of 1.5 and the areal cutoff of 3.5, the sensitivity, specificity, positive predictive value, and negative predictive value for MALT lymphoma were 96.97%, 70.37%, 80.00%, and 95.00%, respectively. In conclusion, through objective measurement of the marginal zone, suspected cases of MALT lymphoma requiring a molecular clonality test can be effectively selected.

Tissue damage caused by various stimuli under certain conditions, such as biological and environmental cues, can actively induce systemic and/or local immune responses. Therefore, understanding the immunological perspective would be critical to not only regulating homeostasis of organs and tissues but also to restrict and remodel their damage. Lungs serve as one of the key immunological organs, and thus, in the present article, we focus on the innate and adaptive immune systems involved in remodeling and engineering lung tissue. Innate immune cells are known to react immediately to damage. Macrophages, one of the most widely studied types of innate immune cells, are known to be involved in tissue damage and remodeling, while type 2 innate lymphoid cells (ILC2s) have recently been revealed as an important cell type responsible for tissue remodeling. On the other hand, adaptive immune cells are also involved in damage control. In particular, resident memory T cells in the lung prevent prolonged disease that causes tissue damage. In this review, we first outlined the structure of the respiratory system with biological and environmental cues and the innate/adaptive immune responses in the lung. It is our hope that understanding an immunological perspective for tissue remodeling and damage control in the lung will be beneficial for stakeholders in this area.

Scrub typhus, a mite-borne infectious disease, is caused by Orientia tsutsugamushi. Despite many attempts to develop a protective strategy, an effective preventive vaccine has not been developed. The identification of appropriate Ags that cover diverse antigenic strains and provide long-lasting immunity is a fundamental challenge in the development of a scrub typhus vaccine. We investigated whether this limitation could be overcome by harnessing the nanoparticle-forming polysorbitol transporter (PST) for an O. tsutsugamushi vaccine strategy. Two target proteins, 56-kDa type-specific Ag (TSA56) and surface cell Ag A (ScaA) were used as vaccine candidates. PST formed stable nano-size complexes with TSA56 (TSA56-PST) and ScaA (ScaA-PST); neither exhibited cytotoxicity. The formation of Ag-specific IgG2a, IgG2b, and IgA in mice was enhanced by intranasal vaccination with TSA56-PST or ScaA-PST. The vaccines containing PST induced Ag-specific proliferation of CD8+ and CD4+ T cells. Furthermore, the vaccines containing PST improved the mouse survival against O. tsutsugamushi infection. Collectively, the present study indicated that PST could enhance both Ag-specific humoral immunity and T cell response, which are essential to effectively confer protective immunity against O. tsutsugamushi infection. These findings suggest that PST has potential for use in an intranasal vaccination strategy.

Routine stress monitoring in daily life can predict potentially serious health impacts. Effective stress monitoring in medical and healthcare fields is dependent upon accurate determination of stress-related features. In this study, we determined the optimal stress-related features for effective monitoring of cumulative stress. We first investigated the effects of short- and long-term stress on various heart rate variability (HRV) features using a rodent model. Subsequently, we determined an optimal HRV feature set using support vector machine-recursive feature elimination (SVM-RFE). Experimental results indicate that the HRV time domain features generally decrease under long-term stress, and the HRV frequency domain features have substantially significant differences under short-term stress. Further, an SVM classifier with a radial basis function kernel proved most accurate (93.11%) when using an optimal HRV feature set comprising the mean of R-R intervals (mRR), the standard deviation of R-R intervals (SDRR), and the coefficient of variance of R-R intervals (CVRR) as time domain features, and the normalized low frequency (nLF) and the normalized high frequency (nHF) as frequency domain features. Our findings indicate that the optimal HRV features identified in this study can effectively and efficiently detect stress. This knowledge facilitates development of in-facility and mobile healthcare system designs to support stress monitoring in daily life.

A novel biochar-coated zero-valent iron [Fe(0)], which was synthesized with rice straw and Fe(0), was applied to remove nitro explosives (2,4,6-trinitrotoluene and hexahydro-1,3,5-trinitro-1,3,5-triazine) and halogenated phenols (2,4-dibromophenol and 2,4-difluorophenol) from contaminated waters. Due to the presence of biochar on the outside, the removal of nitro explosives and halogenated phenols was significantly enhanced via sorption. The sorbed contaminants were further transformed into reductive products, indicating that the inner Fe(0) played the role of a reductant in the biochar-coated Fe(0). Compared to direct reduction with Fe(0), the reductive transformation with biochar-coated Fe(0) was markedly enhanced, suggesting that the biochar in biochar-coated Fe(0) may act as an electron transfer mediator. Further experiments showed that the surface functional groups of biochar were involved in the catalytic enhancement of electron transfer. Our results suggested that biomass could be used to synthesize a novel sorbent and catalyst for treating redox-sensitive contaminants in natural and engineered systems.

In this study, we tested the hypothesis that chronic stress has cumulative effects over time on heart rate variability (HRV) and physiological responses in a rodent model of chronic mild stress. Rats were exposed to either short-term (2weeks) or long-term (4weeks) stress, followed by a 1-week recovery period. Controls were normally housed rats that did not undergo the stress procedure. For electrocardiogram recordings, transmitters were implanted in all rats 10days before the onset of the experiment to allow recovery from surgery. To investigate physiological responses, body weight, temperature, sucrose preference, and serum corticosterone levels were measured weekly. Rats exposed to both short- and long-term stress showed significant reductions in body weight, which did not normalize after the recovery period. A significant difference was observed between short- and long-term stress groups in serum corticosterone levels, with long-term stress significantly increasing serum corticosterone levels, which remained elevated after the recovery period (P<0.05). HRV analysis indicated that long-term stress significantly decreased time-domain indexes, whereas significantly increased frequency-domain indexes were observed in the low-frequency range (0.1-1Hz). These results may represent dysfunction in parasympathetic/vagal modulation with hyperactivation of the sympathetic nervous system after long-term exposures to stress. In addition, prolonged Q-to-T wave (QT) intervals were observed in rats exposed to long-term stress, which did not return to baseline levels after the recovery period. These findings are consistent with the view that chronic stress is associated with cardiac autonomic disorders and emphasize the importance of monitoring stress in our daily life since the effects of even mild stress may be cumulative.

We studied the electrochemical characteristics of tin dioxide (SnO2) recovered from waste catalyst material which had been previously used in a polymer synthesis reaction. In order to improve the electrochemical performance of the SnO2 anode electrode, we synthesized a nanocomposite of recovered SnO2 and commercial iron oxide (Fe2O3) (weight ratio 95:5) using a solid state method. X-ray diffraction (XRD) and field emission scanning electron microscopy (FE-SEM) analyses revealed an additional iron oxide phase within a porous nanocomposite architecture. The electrochemical characterizations were based on galvanostatic charge-discharge (CD) curves, cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). In the first discharge, the capacity of the SnO2-Fe2O3 nanocomposite was 1700 mAh g(-1), but was reduced to about 1200 mAh g(-1) in the second discharge. Thereafter, a discharge capacity of about 1000 mAh g(-1)was maintained up to the 20th cycle. The SnO2-Fe2O3 nanocomposite showed better reversible capacities and rate capabilities than either the recovered SnO2 or commercial Fe2O3 nanoparticle samples.
